Emerging role for R-loops in chromosome segregation
Munira A Basrai, Prashant K Mishra
doi:10.1098/rsob.260090Abstract
R-loops are evolutionarily conserved three-stranded nucleic acid structures formed when a nascent RNA transcript hybridizes with its complementary DNA strand, thereby exposing the non-template strand as single-stranded DNA. Traditionally viewed as a by-product of transcription, R-loops are now recognized as critical regulators of diverse cellular processes, including gene expression, DNA replication, damage and repair, telomere length regulation, mitochondrial function and chromosome segregation. Here, we review studies that define the role of R-loops in ensuring faithful chromosome segregation during cell division and discuss how defects in R-loop dynamics lead to kinetochore (centromeric DNA and associated proteins) dysfunction and chromosomal instability. Taken together, these studies suggest that R-loops function as double-edged swords necessary for accurate chromosome segregation under physiological conditions, yet are deleterious if not resolved and improperly regulated. Given that R-loops have been associated with various diseases, targeting R-loop dynamics may offer novel therapeutic strategies for the treatment of diseases such as cancer.

How to check this record
- Subscribe a DKG node to the context graph above and fetch the asset by its UAL: a UAL alone does not name its graph. The node returns the title, authors, PDF link, submitter, signature and deadline.
- Check the signature. It's EIP-712 typed data with the domain
{ name: "labstrail", version: "1", chainId: 84532 }and the typeSubmission(string cid, string contextGraph,uint256 deadline). A paper published before LabsTrail was renamed is signed with the name"verisci"instead. Any EIP-712 library recovers the signer to compare with the submitter; a smart-contract wallet is checked on chain with ERC-1271. - Download the PDF by its CID. An IPFS client checks every block against its hash, so a changed file can't come back under this CID. To recompute the CID from a local copy, use Pinata's v1 import settings: CIDv1, raw leaves, 256 KiB chunks.
- Open the asset token on Basescan. It's an ERC-721 token in OriginTrail's DKGKnowledgeAssets contract, minted to LabsTrail's node, and its page shows when. The merkle root of the asset's statements is anchored with it, so any node can check that the content still matches.
Ratings
Emerging role for R-loops in chromosome segregation scores 82/100 for reproducibility readiness on LabsTrail. How the score works
